When performing a standard plate count, plates that contain 30 - 300  Bacterial colonies are selected for counting with the Quebec colony counter.

The countable plate has between 30 and 300 colonies. More than 300 Bacterial colonies  would be difficult to count, and less than 30 Bacterial colonies  is too small a sample size to present an accurate representation of the original sample.

The Bacterial colonies is isolated with a colony-picking tool such as a toothpick, inoculation loop, or pipette tip. It is then inoculated in a solid or liquid cell culture medium where it is incubated overnight to encourage replication. The general formula to calculate CFU/ml is well known: The number of microbes/ml = number of Bacterial colonies  x dilution of the sample. Between 0.3 ml and 0.5 ml of a suspension of indicator bacteria is required for each plate.
